protein CrDAT mô tế bào cụ thể thực vật Tuy nhiên, yếu tố cis nhân tố trans có vai trò cụ thể với biểu gen CrDAT thực vật chuyển gen cần tiếp phân tích làm sáng tỏ Cấu trúc 35S-CrDAT-cmyc chuyển thành công vào mô thuốc N tabacum K326 nhờ A tumefaciens, tạo thuốc chuyển gen Sự có mặt hợp gen chuyển CrDAT vào hệ gen thuốc chuyển gen kiểm tra PCR Southern blot Protein tái tổ hợp CrDAT có trọng lượng phân tử khoảng 51 kDa biểu dòng thuốc chuyển gen Kết biến nạp đánh giá hoạt động cấu trúc 35SCrDAT-cmyc thuốc chuyển gen sở cho viêc̣ điều khiển biểu mạnh gen CrDAT dừa cạn, ta ̣o dòng dừa cạn chuyển gen có hàm lượng vincristine vinblastine cải thiện 54 KẾT LUẬN VÀ ĐỀ NGHỊ Kết luận 1.1 Gen CrDAT phân lập từ dừa cạn tách dòng phân tử xác định trình tự nucleotide Gen CrDAT (cDNA) có kích thước 1320 bp, mã hóa cho 439 amino acid 1.2 Vector chuyển gen thực vâ ̣t pBI121-CrDAT thiết kế chuyển vào hệ gen thuốc dừa cạn chuyển gen biểu protein tái tổ hợp CrDAT có kích thước khoảng 51 kDa 1.3 Môi trường MS bổ sung sucrose 30 g/l; agar 10 g/l; BAP 1,0 mg/l IBA 0,6 mg/l; nước dừa 100 ml/l; pH = 5,8 thích hợp cho phát sinh chồi sự sinh trưởng chồi từ đoạn thân mang mắt chồi bên Môi trường MS bổ sung sucrose 30 g/l; agar 10 g/l; BAP 0,5 mg/l IBA 0,4 mg/l; nước dừa 100 ml/l; pH = 5,8, thích hợp cho phát sinh chồi sinh trưởng chồi từ nách mầm Đề nghị Tiếp tu ̣c chuyển gen DAT vào dừa cạn phân tích
